Immunohistochemistry-Paraffin: Nbs1 Antibody [NB100-143] - FFPE human testis using Nbs1 antibody [NB100-143] at 1:200 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Staining was performed by Histowiz.

Western Blot: Nbs1 Antibody [NB100-143] - Analysis of HeLa whole cell lysate [NB800-PC1] using rabbit polyclonal NBS1 antibody [NB100-143]. Observed Molecular Weight at ~95 kDa.

Cell lysates were prepared and separated on a 12% gel by SDS-PAGE. Protein was transferred to PVDF membrane and blocked in 5% non-fat milk. The membrane was then probed with 1 ug/ml anti-p62/SQSMT1 in 1% milk and dectected with an anti-rabbit HRP secondary antibody using chemiluminescence. Note the upregulation of p62 (arrowhead) in response to chloroquine treatment and the blockage of autophagy.

Immunohistochemistry-Paraffin: Beclin 1 Antibody [NB110-87318] - Analysis of a FFPE tissue section of human breast cancer using Beclin 1 antibody (Lot 71917) at 1:300 dilution. The primary antibody bound to Beclin 1 antigen in the tissue section was detected using a HRP labeled secondary antibody and DAB reagent. Nuclei of the cells were counterstained with hematoxylin. Cytoplasmic staining can be seen in the cancer as well as the stromal cells. The signal was strongest in a subset of stromal cells which appears to be the cancer associated fibroblasts.

Western Blot: Beclin 1 Antibody [NB110-87318] - Basal levels of autophagy in the hypothalamus of BACHD mice. The expression levels of selected autophagy markers were analyzed by Western blot in hypothalamic tissue from BACHD mice at 2, 6 and 12 months of age and compared with gender- and age-matched controls (n=3-6/group). The data showed a substantial maintenance of the autophagy flux, despite the presence of a metabolic phenotype in the BACHD mice already at early time points. CD34 was detected in immersion fixed paraffin-embedded sections of mouse Thymus using Rat Anti-Mouse CD34, Monoclonal Antibody (Catalog #NB600-1071) at 1:500 dilution at 37 ° Celsius for 2 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH).Tissue was stained using the Alexa Fluor™ 647 Goat anti-Rat IgG Secondary Antibody at 1:200 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR647RT) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to the membrane. Protocol available in COMET™ Panel Builder.

Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a FFPE tissue section of mouse small intestine using rat anti-mouse CD34 (clone MEC 14.7) at 1:100 dilution. The signal was developed using HRP-conjugated anti-rat secondary with DAB reagent which followed counterstaining of nuclei using hematoxylin. This antibody specifically labelled the endothelial cells in blood vessels located primarily in the sub-mucosa, and of that of the mucosa muscularis and the mucosal lacteal.

Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a FFPE tissue section of human renal cancer xenograft using CD34 antibody clone MEC 14.7 at 1:100 dilution. The signal was developed using HRP-conjugated secondary antibody and DAB reagent followed by counterstaining of nuclei with hematoxylin. Notably, the antibody specifically stained the endothelial cells of the tumor vasculature which is originating from the host animal cells (mouse). This observation was verified by including a negative control in this validation testing (see the IHC image of human renal cancer tissue section with negative signal for CD34).

Flow Cytometry: CD34 Antibody (MEC 14.7) [NB600-1071] - An intracellular stain was performed on Raw264.7 cells with CD34 Antibody [MEC 14.7] NB600-1071 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rat IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 488 (SA5-10018, Thermo Fisher).
